拟南芥Mo敏感变异株的筛选和表型分析

摘 要:

本文以EMS诱变处理野生型拟南芥Col-0获得的M2代种子为试验材料, 通过在不含钼(-Mo)和含170 nmol•L-1钼(+Mo)的培养基上进行反复筛选培养, 根据拟南芥根系在2种培养基上的生长差异筛选出31株Mo敏感变异株。并对变异株2-4的表型特征及鲜重和Mo含量进行了分析。结果显示, 在-Mo培养基中变异株2-4的生长明显受到抑制, 且叶鲜重、根鲜重和Mo含量均低于野生型。而在+Mo培养基中变异株2-4叶鲜重、根鲜重与野生型的差异不明显, 且都高于在-Mo培养基中的。随着钼含量的增加, 变异株2-4矮小、叶片失绿和叶缘卷曲的症状不断恢复。当钼含量达到170 nmol•L-1时, 变异株2-4的生长情况与野生型相比没有明显差异。这说明筛选出的2-4植株是一种Mo敏感型变异株。

Screening and Phenotypic Analysis of Mo-Sensitive Mutants in Arabidopsis thaliana

Abstract:

In this study, M2 generation seeds from wild-type Arabidopsis thaliana Col-0 treated with ethylmethane sulfonate (EMS) were used as experimental materials, which were cultured on the no-molybdenum (-Mo) and 170 nmol•L-1 molybdenum (+Mo) media for repeated screening. According to the growth status of roots in two media, 31 plantlets of Arabidopsis Mo-sensitive mutants were selected. Subsequently, the phenotype, fresh weight and Mo content of mutant 2-4 were analyzed. The results showed that the growth of mutant 2-4 was significantly inhibited, and fresh weights and Mo contents of leaves and roots in mutant 2-4 were lower than those of wild type Col-0 on the -Mo medium. But fresh weights of leaves and roots of mutant 2-4 and wild type Col-0 were not obviously different on the +Mo medium, and fresh weights and Mo contents were far higher than those on the -Mo medium.With the Mo content in the medium increasing, the symptoms of dwarf, leaf chlorosis and margin of leaf curl of mutant 2-4 recovered continuously. When the Mo content reached to 170 nmol•L-1, the growth of mutant 2-4 had no obvious differences with the wild-type. These results showed that screened 2-4 plant was a Mo-sensitive mutant.
